The TOP assay is a powerful technique used to identify the hidden or unknown fraction of PFAS compounds in environmental samples. By treating water, soil, sediment, and biota with persulfate, hydroxide, and heat to induce hydroxyl radical oxidation, polyfluorinated precursors are converted into more persistent perfluorinated forms such as PFOA and PFOS. After oxidation, PFAS compounds are extracted and quantified using isotope dilution LC/MS/MS.
